生化机理

盐活性核酸酶（SAN）是一种通用的、非特异性核酸内切酶，可切割双链和单链 DNA 和 RNA。SAN 在高于中性 pH 时具有活性。但是，与其他核酸酶不同，它在高 pH、高盐浓度下具有最佳活性。这些特性使 SAN 成为从细胞提取物和蛋白质样品中去除 DNA 的理想选择。DNA 完全降解后的最终产物由 5 个核苷酸的寡核苷酸组成这种酶降解 DNA 和 RNA 的比率为 10:1。 SAN 在 10-40°C 的温度范围内具有很高的活性。活性的最佳 NaCl 浓度为 0.5 M。但是，SAN 在低盐缓冲液中具有活性。不建议将 1 M NaCl 用于 SAN。活性需要 Mg2 +（>1 mM）。pH 的工作范围高于中性 pH，最佳范围在 8.5-9.5 之间物理形式：溶于 25 mM Tris-HCl（pH 7.5）、5 mM MgCl2、0.5 M NaCl、0.01％ Triton 和 50％（v/v）甘油的溶液单位定义：一个单位（U）定义为在 37°C 下，30 分钟内，在含 5 mM MgCl2 和 500 mM NaCl 的 25 mM Tris-HCl（pH 8.5，25ºC）中使用 50 μg/ml 小牛胸腺 DNA 在 260 nm 处的吸光度增加量。
