阿拉丁生产的T4 Gene 32 Protein (ssDNA结合蛋白),也称T4 gp32 protein,T4 gp32蛋白或T4噬菌体gp32蛋白,是一种由噬菌体T4基因组的基因32编码的单链DNA (ssDNA)结合蛋白。T4 Gene 32 Protein为T4噬菌体DNA复制和修复所必需。在T4噬菌体DNA复制过程中,T4 gp32结合并稳定暂时形成的单链DNA区域。在使用电子显微镜观察细胞内DNA结构的时候,该蛋白也被广泛用于稳定和标记单链DNA区域。此外,T4 gp32还被报道可以促进限制性内切酶和T4 DNA聚合酶活性,在RT-PCR过程中提高逆转录酶效率,提高PCR产物产量。T4 gp32 protein可用于重组酶聚合酶扩增(recombinase polymerase amplification, RPA)。RPA主要包括以下几个组分:重组酶(recombinase) T4 UvsX,重组酶载入因子(recombinase loading factor) T4 UvsY,Bsu'DNA聚合酶大片段以及单链DNA结合蛋白(single-strand binding protein) (如T4 gp32 Protein)等。在ATP参与的情况下,UvsX与引物相结合形成复合物,扫描DNA模板,寻找到与引物配对的核酸序列。一旦引物被定位到了DNA模板的同源序列,可以直接引发链交换反应形成D-Loop结构:ATP水解产生ADP,UvsX被gp32取代而从DNA模板上解离下来,gp32作为单链结合蛋白可以稳定在被置换的DNA链的D-Loop结构中,防止引物解离。UvsX被解离后,引物3'末端暴露并被"Bsu DNA聚合酶识别。随后聚合酶与DNA模板配对并进行引物的延伸和模板的扩增。重组酶载入因子T4 UvsY和拥挤试剂(crowding reagent) Carbowax20M的加入有利于UvsX与引物的结合,即有利于重组酶UvsX的载入。 用途 : 稳定或标记ssDNA;在RT-PCR过程中,增加反转录产物的产量;增强体外DNA的合成;与T4 DNA Polymerase和T4 DNA Ligase一起用于位点特异的突变实验;从含有腐植酸的样本如土壤中扩增细菌或真菌基因组时,可提高PCR产物的产量和特异性(当PCR体系中包含T4 gp32蛋白时(2.5μg/100μl),腐植酸的抑制作用可被降低7倍);促进限制性内切酶的消化反应。 来源 : 大肠杆菌表达和纯化而得的T4 噬菌体基因32的重组蛋白。 酶储存溶液 : 20mM Tris-HCl (pH8.0), 100mM NaCl, 0.5mM DTT, 1mM EDTA, 50% (v/v) Glycerol.10X T4 Gene 32 Protein Buffer:200mM Tris-Acetate (pH7.9 at 25ºC), 500mM Potassium Acetate, 100mM Magnesium Acetate, 10mM DTT. 失活或抑制 : 65℃孵育20min。 注意事项 : 本产品仅限于专业人员的科学研究用,不得用于临床诊断或治疗,不得用于食品或药品,不得存放于普通住宅内。为了您的安全和健康,请穿实验服并戴一次性手套操作。

Single-stranded DNA-binding protein that participates in viral DNA replication, recombination, and repair (Probable). Coats the lagging-strand ssDNA as the replication fork advances.Stimulates the activities of viral DNA polymerase and DnaB-like SF4 replicative helicase, probably via its interaction with the helicase assembly factor. Stimulates the unwinding activity of UvsW helicase, inhibits it DNA winding activity. Together with DnaB-like SF4 replicative helicase and the helicase assembly factor, promotes pairing of two homologous DNA molecules containing complementary single-stranded regions and mediates homologous DNA strand exchange. Promotes also the formation of joint molecules. mRNA specific autogenous translational repressor.
