本公司生产的 T4 RNA 连接酶 2 截短型(突变型)(T4 Rnl2tr KQ, or T4 Rnl2tr R55K, K227Q),是 T4 RNA 连接酶 2 截短型的 R55K 和 K227Q 双点 突变体,可以大大减少T4 RNA Ligase 2, truncated的对于RNA的非特异性连接(RNA的串联或自连成环),并能保持和T4 RNA Ligase 2, truncated 相近的酶活性。T4 RNA Ligase 2, truncated KQ 是在 T4 RNA Ligase 2, truncated 的基础上将 227 位的赖氨酸和 55 位的精氨酸分别突变为谷氨 酰胺和赖氨酸,K227Q 突变后大大降低了 T4 RNA Ligase 2, truncated 该酶所含有的轻微的从 5’预腺苷酰化的 DNA 接头(AppDNA)将腺苷基团转 移到 RNA 5’磷酸基团同时形成 5’磷酸化 DNA 接头(pDNA)的活性,从而减少了 T4 RNA Ligase 2, truncated 导致的 RNA 非特异性连接(RNA 串联 或自连成环),但 T4 RNA Ligase 2, truncated K227Q 的连接酶活性有显著下降。在 T4 RNA Ligase 2, truncated K227Q 基础上进一步突变 R55K 后,酶的连接活性提升到了与野生型 T4 RNA Ligase 2, truncated 的连接活性相似的状态。T4 RNA Ligase 2, truncated KQ 与 T4 RNA Ligase 2, truncated具有类似的连接 ssRNA和AppDNA的效果,但T4 RNA Ligase 2, truncated能够使 AppDNA去腺苷酰化,形成pDNA,而T4 RNA Ligase 2, truncated KQ 对 AppDNA 不具有去腺苷酰化活性。T4 RNA Ligase 2, truncated KQ 常用于 microRNA (miRNA)等 3’端为羟基的单链 RNA 在 克隆、高通量测序建库或 PCR 检测等时,在 3’羟基端添加 5’端预腺苷酰化的 DNA 或 RNA 接头。T4 RNA Ligase 2, truncated KQ 只能利用 5’端预 腺苷酰化的单链 DNA 或 RNA 作为 3’端接头,因此可以大大降低连接反应的背景,通常是小 RNA 等建库时的优先选择。T4 RNA Ligase 2, truncated KQ 连接时不需要 ATP,但需要 5’端预腺苷酰化(也称预腺苷化、腺苷酰化或腺苷化)底物。 纯度(Purity):无 DNA 内切酶和外切酶活性,无 RNA 酶活性,无蛋白酶活性。 组分和说明 T750875
生化机理
Repairs 3'-OH/5'-PO4 nicks in duplex RNA or RNA:DNA hybrid in which the broken 3'-OH strand is RNA (Probable) .The nick ligation reaction entails three nucleotidyl transfer steps. In the first step, the RNA ligase reacts with ATP in the absence of nucleic acid to form a covalent ligase-AMP intermediate and release pyrophosphate. In step 2, the ligase-AMP binds to the nicked duplex nucleic acid and transfers the adenylate to the 5'-PO4 terminus to form an adenylylated nicked intermediate. In step 3, the RNA ligase directs the attack of the nick 3'-OH on the 5'-phosphoanhydride linkage, resulting in a repaired 3' - 5' phosphodiester and release of AMP.
