生化机理

由活化的 CD4 阳性辅助 T 细胞产生的细胞因子，其次是活化的 CD8 阳性 T 细胞和自然杀伤（NK）细胞，在免疫反应和耐受中起着关键作用。与由高亲和力三聚体 IL-2R（IL2RA/CD25、IL2RB/CD122 和 IL2RG/CD132）或低亲和力二聚体 IL-2R（IL2RB 和 IL2RG）组成的受体复合物结合。与受体的相互作用会导致 IL-2R 亚基的寡聚化和构象变化，从而产生下游信号，首先是 JAK1 和 JAK3 的磷酸化。反过来，JAK1 和 JAK3 又使受体磷酸化，形成一个对接位点，导致包括 STAT5 在内的多个底物磷酸化。这一过程会激活多种途径，包括 STAT、磷酸肌醇-3-激酶/PI3K 和丝裂原活化蛋白激酶/MAPK 途径。作为一种 T 细胞生长因子，还能提高 NK 细胞的细胞溶解活性。促进活化 B 细胞的强烈增殖，进而促进免疫球蛋白的产生。通过控制维持免疫耐受所需的调节性 T 细胞的存活和增殖，在调节适应性免疫系统中发挥关键作用。此外，它还参与效应 T 细胞亚群（包括 Th1、Th2、Th17 以及记忆 CD8 阳性 T 细胞）的分化和稳态。

生物活性

Measured in a cell proliferation assay using CTLL‑2 mouse cytotoxic T cells. The ED50 for this effect is 0.0300-0.250 ng/mL. The specific activity of recombinant human IL-2 is >5.00 x 10^6 IU/mg, which is calibrated against the human IL-2 WHO International Standard (NIBSC code: 86/500).
