生化机理

主要的免疫调节细胞因子，作用于免疫系统中的许多细胞，具有深远的抗炎功能，可限制炎症对组织造成的过度破坏。从机理上讲，IL10 与其由 IL10RA 和 IL10RB 组成的异源四聚体受体结合，导致 JAK1 和 STAT2 介导的 STAT3 磷酸化。反过来，STAT3 会转位到细胞核，并在细胞核中驱动抗炎介质的表达。靶向巨噬细胞和单核细胞等抗原递呈细胞（APC），抑制它们释放促炎细胞因子，包括粒细胞-巨噬细胞集落刺激因子/GM-CSF、粒细胞集落刺激因子/G-CSF、IL-1 alpha、IL-1 beta、IL-6、IL-8 和 TNF-alpha。还能通过减少 MHC II 级和协同刺激分子的表达来干扰抗原递呈，从而抑制它们诱导 T 细胞活化的能力。此外，它还能通过重新规划包括 mTOR 信号在内的重要代谢途径来控制巨噬细胞的炎症反应。

生物活性

Measured in a cell proliferation assay using MC/9‑2 mouse mast cells. The ED50 for this effect is 0.0750-0.750 ng/mL. The specific activity of recombinant human IL-10 is >2.50 x 10^6 units/mg, which is calibrated against the human IL-10 WHO Reference Reagent (NIBSC code: 93/722).
