生化机理

Chromogranin A（CgA）又称垂体分泌蛋白 I（SP-I），是粒蛋白家族中的一种调节分泌蛋白。CgA 具有粒蛋白家族的几个共同特征：酸性等电点、结合钙离子的能力、形成聚集体的能力和多个二碱基裂解位点。成熟的人类 CgA 有 439 个氨基酸（aa），包含 10 个二碱基蛋白水解裂解位点，能够产生多个较小的肽，每个肽都具有独特的功能。成熟的人类 CgA 与小鼠和大鼠 CgA 有 63% 的 aa 序列相同性。CgA 只在大多数正常和肿瘤性神经内分泌细胞的分泌型致密核心颗粒中表达。在神经内分泌肿瘤和非神经内分泌肿瘤患者体内均检测到 CgA 水平升高，因此 CgA 是诊断肿瘤和监测肿瘤进展/消退的重要血清学标志物。在小鼠模型中已经证实，含有 C 端区域的全长 CgA 能抑制血管生成和肿瘤生长。此外，CgA 还能与 Secretogranin III 结合，调节分泌颗粒的生物生成。
