生化机理

泛素结合酶 H5b（UbcH5b）又称泛素结合酶 E2D 2（UBE2D2），是泛素结合（E2）酶家族中一个广泛表达的成员。该蛋白的预测分子量为 16.5 kDa。UbcH5b/UBE2D2 同时定位于细胞核和细胞质。人类蛋白质与小鼠和大鼠的直向同源物分别具有 100% 和 92% 的氨基酸序列相同性。该酶的 E2 催化核心结构域含有一个活性位点半胱氨酸残基，该残基是与泛素形成硫酯键所必需的。UbcH5b/UBE2D2 能够介导通过 Lys11、Lys48 或 Lys63 链接的泛素链的形成。UbcH5b/UBE2D2 与 SCF(Fbxw2) 和 MDM2/HDM2 泛素连接酶（E3）合作，分别介导转录因子 GCM1 和 p53 的泛素化和降解。与 UBE2N/Ubc13 一起，UbcH5b/UBE2D2 可能在 MHC I 类分子的内吞和溶酶体内降解中发挥作用。据报道，UbcH5b/UBE2D2 对 TRIM5-α 的非蛋白水解泛素化作用可阻止 HIV 的逆转录。从病理学角度看，UBE2D 家族成员可能是镉诱导肾毒性过程中镉的关键靶标。此外，UbcH5b/UBE2D2 的过度表达还与炎症性肠病有关。
