生化机理

CEACAM5 又称 CEA 或 CD66e，属于免疫球蛋白超家族中的 CEACAM 大亚家族。CEACAM5 主要由上皮细胞表达，合成为一种分子量为 180 kDa 的糖蛋白，其中 60% 为碳水化合物。CEACAM5 的 N 端含有一个 Ig 样 V 型结构域，其后是六个 Ig 样 C2- 型结构域和一个 GPI 锚点，以同源二聚体形式存在。CEACAM5 和 CEACAM6 在许多癌症中过表达，与粘附和侵袭有关。CEACAM5 可通过同型和异型相互作用介导细胞-细胞粘附。CEACAM5 是一种同型细胞间粘附分子，在肿瘤组织中的表达水平高于相应的正常组织，因此被广泛用作肿瘤标志物。研究还表明，CEACAM5 可抑制细胞分化，从而增加肿瘤的致病性。此外，CEACAM5 还被确定为大肠杆菌 Dr 系列粘附素的宿主受体，大肠杆菌 Dr 粘附素的结合会导致 CEACAM5 同源二聚体的解离。
