生化机理

凝血因子 III/组织因子（TF）又称凝血活酶和 CD142，是一种存在于多种类型细胞中的完整膜蛋白。它是凝血因子 VII 的蛋白质辅助因子/受体，凝血因子 VII 在肝脏中合成，在血浆中循环。与 TF 结合后，非活性因子 VII 会迅速转化为活性 VIIa。VIIa 和 TF 形成的 1:1 复合物可启动凝血途径，还具有重要的非凝血功能，如血管生成。TF 合成为 295 个氨基酸的前体，由信号肽（残基 1 至 32）和成熟链（残基 33 至 295）组成。作为 I 型膜蛋白，它包含跨膜区（残基 252 至 274）和胞质尾（残基 275 至 295）。纯化的 rhTF 与外结构域（残基 34 至 251）相对应，在《活性测定协议》所述条件下可有效激活热溶解酶处理的重组人凝血因子 VII（目录编号 2338-SE）。
