生化机理

酪氨酸蛋白激酶是细胞因子 KITLG/SCF 的细胞表面受体，在细胞存活和增殖、造血、干细胞维持、配子生成、肥大细胞发育、迁移和功能以及黑色素生成的调控中发挥着重要作用。与 KITLG/SCF 结合后，KIT 可激活多种信号通路。磷酸化 PIK3R1、PLCG1、SH2B2/APS 和 CBL。通过磷酸化磷脂肌醇 3- 激酶的调节亚基 PIK3R1，激活 AKT1 信号通路。活化的 KIT 还可通过 GRB2 和激活 RAS、RAF1 及 MAP 激酶 MAPK1/ERK2 和/或 MAPK3/ERK1 传递信号。促进 STAT 家族成员 STAT1、STAT3、STAT5A 和 STAT5B 的活化。PLCG1 的激活会导致细胞信号分子二酰甘油和 1,4,5-三磷酸肌醇的产生。KIT 信号通过蛋白磷酸酶以及受体的快速内化和降解进行调节。活化的 KIT 会促进蛋白磷酸酶 PTPN6/SHP-1 和 PTPRU 以及转录因子 STAT1、STAT3、STAT5A 和 STAT5B 的磷酸化。促进 PIK3R1、CBL、CRK（异构体 Crk-II）、LYN、MAPK1/ERK2 和/或 MAPK3/ERK1、PLCG1、SRC 和 SHC1 的磷酸化。
