生化机理

NME1又称核苷二磷酸激酶A（NDK-A）或NM23-H1，属于NDK家族。众所周知，NM23-H1 在许多肿瘤细胞中具有抑制转移的活性。最近的研究表明，与 NM23-H1 相互作用的蛋白能介导细胞增殖，也可能是转移抑制活性的调节因子。与 NM23-H1 相互作用的蛋白质可分为三类。第一类蛋白可归为 NM23-H1 的上游激酶，如 CKI 和 Aurora-A/STK15。第二类蛋白在 Erk 信号级联中充当下游效应器，调节特定基因转录、GTP 结合蛋白功能和信号转导。第三类蛋白质可归类为 NM23-H1 的双向影响结合伙伴。因此，NM23-H1 与结合伙伴的相互作用对涉及转移和肿瘤发生的生化特征具有影响。NDKA 在人死后脑脊液（CSF）中增加，而脑脊液是脑部全面损伤的模型，这表明在脑脊液中，更重要的是在血浆中测量 NDKA 可作为中风的生物标志物。此外，NM23-H1 能显著减少转移，而不影响原发肿瘤的大小，是第一个被发现的转移抑制基因。
