生化机理

酪氨酸蛋白激酶是成纤维细胞生长因子的细胞表面受体，在调节细胞增殖、分化和迁移以及调节脂质代谢、胆汁酸生物合成、葡萄糖摄取、维生素 D 代谢和磷酸盐稳态中发挥作用。在 FGF19 的作用下，需要正常下调胆汁酸合成的限速酶 CYP7A1 的表达。磷酸化 PLCG1 和 FRS2。配体结合会激活多个信号级联。激活 PLCG1 会导致细胞信号分子二酰甘油和 1,4,5-三磷酸肌醇的产生。FRS2 磷酸化会引发 GRB2、GAB1、PIK3R1 和 SOS1 的招募，并介导 RAS、MAPK1/ERK2、MAPK3/ERK1 和 MAP 激酶信号通路以及 AKT1 信号通路的激活。促进依赖于 SRC 的基质蛋白酶 MMP14 磷酸化及其溶酶体降解。FGFR4 信号通过受体内化和降解而下调；MMP14 促进 FGFR4 的内化和降解。导致组成激酶激活或损害正常 FGFR4 失活的突变会导致异常信号传导。翻译后：N-糖基化。糖链在高尔基体中的完全成熟对于与 FGF19 的高亲和力相互作用至关重要。泛素化。未完全糖基化时会被蛋白酶体降解。自身磷酸化。FGF 家族成员与硫酸肝素蛋白多糖或肝素结合，促进受体二聚化和酪氨酸残基上的自动磷酸化。自动磷酸化在二聚体中的两个 FGFR 分子之间反式发生。
