生化机理

乙醇酸氧化酶是α 羟基酸氧化酶（HAO）超家族的成员，植物和动物中都存在这种酶。它催化 FMN 介导的乙醇酸氧化为乙醛酸，乙醛酸氧化为草酸，同时将氧还原为过氧化氢。辅助因子 FMN 与蛋白质紧密结合，但不是共价连接。在人类和其他脊椎动物中，HAOs 主要存在于肝脏、肾脏和胰腺的过氧物酶体中。人类已发现三种 HAO。HAO-1 在肝脏和胰腺中的表达量最高，对乙醇酸等二碳底物的活性最强。HAO-2 在肝脏和肾脏中表达，对长链 alpha 羟基酸底物（如 2羟基棕榈酸酯）具有更强的活性。HAO-3 主要在胰腺中表达。最近，HAO 被确认为高草酸尿症（一种草酸钙大量沉积形成肾结石的疾病）的主要成因。
