生化机理

蛋白质 S100-A11，又称 S100 钙结合蛋白 A11、S100A11 和 MLN70，是 S-100 家族的成员。S100A11 在多种组织中广泛表达，位于细胞质、细胞核甚至细胞外围。S100A11 以反平行构象的非共价同源二聚体形式存在。Ca(2+) 与 S100A11 结合会引发构象变化，从而暴露出 S100A11 的疏水裂隙，促进其与靶蛋白的相互作用。作为一种双重细胞生长介质，S100A11 在许多不同类型的肿瘤中充当肿瘤抑制剂或促进剂，并在影响癌细胞增殖方面发挥各自的作用。在细胞核中，S100A11 通过激活 p21（CIP1/WAF1）抑制角质形成细胞的生长，并诱导细胞分化。S100A11 还是乳腺癌的一种新型诊断标志物。翻译后：PRKCA 在 Thr-10 处的磷酸化会显著抑制同源二聚化，并促进与 NCL/nucleolin 的结合，从而诱导核转位。
