生化机理

高移动基团核糖体结合蛋白 1（HMGN1）又称 HMG-14，是非组蛋白染色体蛋白家族的成员，在染色质重塑过程中发挥重要作用。HMGN1 包含一个与核糖体相互作用的 N 端结构域和一个协助染色质展开的 C 端结构域。人类 HMGN1 与小鼠和大鼠 HMGN1 有 80% 的氨基酸序列相同。替代 RNA 处理产生了一种长异构体，在 Met1 之后插入了 45 个氨基酸。HMGN1 在细胞核和细胞质之间的迁移受其多个位点的丝氨酸磷酸化调控。在细胞核中，HMGN1 优先与染色质重塑和基因转录位点结合，从而调节多种基因的表达。它通过直接与双链断裂传感器 PARP-1 结合并激活该传感器，促进 DNA 损伤修复。此外，HMGN1 还是几种警报蛋白之一，这些蛋白通常处于细胞内，但在组织损伤或坏死后会释放出来。在这种情况下，HMGN1 会促进树突状细胞成熟、招募到炎症部位并产生 Th1 炎症细胞因子。这些促炎活动依赖于 HMGN1 与树突状细胞上的 TLR4-MD2 复合物的直接结合。
