生化机理

NAD(P)H：醌受体氧化还原酶 1（NQO1）又称 DT-二磷酸酶，是一种广泛分布的依赖 FAD 的黄蛋白，可促进醌、醌亚胺、硝基芳香族化合物和偶氮染料的 2 电子还原。因此，它能防止醌类化合物的单电子还原，从而防止产生自由基。NQO1 是一种高诱导性酶，受 Keap1/Nrf2/ARE 通路调节。NQO1 水平的升高和降低分别与氧化应激敏感性的降低和升高有关。因此，NQO1 是氧化应激中一种标志性的细胞保护酶。与催化功能无关，NQO1 在调节 p53、p73a 和 p33 的蛋白体降解中发挥作用。NQO1 以一种依赖于 NADH 的方式与 p53 和 p73 发生物理相互作用，并通过一种独立于泛素的途径保护它们免于 20S 蛋白质体降解。
