生化机理

来自大肠杆菌的甘油激酶（glpK）催化甘油的 ATP 依赖性磷酸化，生成 sn-甘油-3-磷酸酯（G3P），这是利用甘油的第一步，也是限制速率的一步。在甘油存在的情况下，glpK 通过与膜结合的甘油促进剂相互作用而受到刺激。在葡萄糖存在的情况下，glpK 的活性受到糖酵解途径中的 1,6-二磷酸果糖（FBP）的异构抑制。在生理条件下，该酶处于活性二聚体和非活性四聚体之间的平衡状态。FBP 与非活性形式结合并使其稳定，从而将甘油代谢途径的使用转移到糖酵解途径。GlpK 是 ATP 酶超家族的成员，该超家族包括肌动蛋白、己糖激酶和热休克蛋白 hsc70。虽然这些蛋白在氨基酸序列和功能上不尽相同，但它们具有相似的三级折叠，而且可能具有相同的催化机制。使用磷酸酶偶联激酶测定法测量了酶的活性。
