生化机理

IL-1 是指两种多效细胞因子，即 IL-1 alpha（IL-1F1）和 IL-1 beta（IL-1F2），它们是不同基因的产物。IL-1 alpha 和 IL-1 beta 是结构相关的多肽，在猪体内大约有 27% 的氨基酸 (aa) 相同。这两种蛋白质都是由多种细胞在炎症因子、感染或微生物内毒素作用下产生的。虽然 IL-1 alpha 和 IL-1 beta 是独立调节的，但它们与相同的受体结合并产生相同的生物效应。IL-1 RI 直接与 IL-1 alpha 或 IL-1 beta 结合，然后与 IL-1 R 辅助蛋白（IL-1 R3/IL-1 R AcP）结合，形成高亲和力的受体复合物，能够进行信号转导。IL-1 RII 对 IL-1 beta 具有高亲和力，但其功能是诱饵受体和 IL-1 beta 活性的负调节剂。IL-1ra 通过阻止 IL-1 alpha 和 IL-1 beta 与 IL-1 RI 相互作用而发挥竞争性拮抗剂的作用。猪 IL-1 beta cDNA 编码 267 aa 的前体。114 aa 的前肽在细胞内被半胱氨酸蛋白酶 IL-1 beta 转换酶（Caspase-1/ICE）裂解，生成活性细胞因子。17 kDa 的成熟猪 IL-1 beta 与犬、棉鼠、马、猫、人、小鼠、大鼠和恒河猴 IL-1 beta 有 63%-70% 的 aa 序列相同性。
