生化机理

IL-1ra 最初是从单核细胞白血病患者的尿液中分离出来的，也是从粘附的单核细胞中纯化出来的。天然存在的全糖基化形式的表观分子量约为 25,000 道尔顿。该蛋白与 IL-1 beta 有 26% 的氨基酸同源性，与 IL-1 alpha 有 19% 的同源性。它会与其中任何一种因子竞争受体结合，但不会与其中任何一种因子相互作用。人 IL-1ra 可与人体细胞上的两种 IL-1 受体（I 型和 II 型）结合。在小鼠体内，IL-1 RII 不与 IL-1ra 结合。重组的非糖基化型 IL-1ra 与天然存在的糖基化型 IL-1ra 一样能阻止 IL-1 与其受体的结合。研究表明，IL-1ra 在体外和体内都能阻断 IL-1 诱导的炎症反应。临床前和临床研究测试了 IL-1ra 在治疗败血症、类风湿性关节炎和慢性骨髓性白血病方面的可能应用。白细胞介素-1 家族促炎细胞因子（如白细胞介素-1β/IL1B 和白细胞介素-1α/IL1A）的抗炎拮抗剂。可防止 IL1 对病原体等一系列先天性刺激因子引发的免疫失调和失控性全身炎症。
