生化机理

IFN gamma 又称 IFNG，是一种分泌型蛋白质，属于 II 型干扰素家族。作为先天性免疫反应的一部分，IFN γ 主要由自然杀伤细胞和自然杀伤 T 细胞产生，而一旦形成抗原特异性免疫，则由 CD4 和 CD8 细胞毒性 T 淋巴细胞效应 T 细胞产生。IFN γ 具有抗病毒、免疫调节和抗肿瘤特性。IFNG 除了具有抗病毒活性外，还具有重要的免疫调节功能，它是巨噬细胞的强效激活剂，对转化细胞具有抗增殖作用，并能增强 I 型干扰素的抗病毒和抗肿瘤作用。IFNG 单体由六个 α-螺旋核心和一个位于 C 端区域的扩展未折叠序列组成。IFN gamma 对抵御病毒和细胞内细菌感染的先天性和适应性免疫以及肿瘤控制至关重要。IFN gamma 的异常表达与一些自身炎症和自身免疫性疾病有关。IFN gamma 在免疫系统中的重要性部分源于其直接抑制病毒复制的能力，最重要的是源于其免疫刺激和免疫调节作用。IFNG 还能促进 NK 细胞的活性。翻译后：蛋白水解加工会产生 C 端异质性，蛋白质可选择以 Gly-150、Met-157 或 Gly-161 结尾。

1. Recombinant human IFN-gamma bioactivity is measured by inhibition of the proliferation of HT-29 cells.The EC50 for this effect is typically 1.41 ng/mL. 2. Immobilized Recombinant Human IFN-gamma protein (rp186559) at 1.0 μg/mL can bind Fontolizumab (anti-IFNG) (Ab175539) with the EC50 of 23.33 ng/mL.
