CD177, also known as NB1, human neutrophil antigen-2a (HNA-2a), and polycythemia rubra vera-1 (PRV-1), is an approximately 50-60 kDa glycoprotein member of the uPAR/CD59/Ly6 superfamily. Mature human CD177 is a 387 amino acid (aa) protein that contains two uPAR/Ly6 domains. It lacks the first two uPAR domains that are present in mouse and rat CD177. Mature human CD177 shares 55% aa sequence identity with common regions of mouse and rat CD177. Alternative splicing of human CD177 generates additional isoforms that are truncated either just before or just after the first uPAR domain. CD177 is expressed on the surface of neutrophils through a glycosylphosphatidylinositol (GPI) anchor. It is nearly absent from neutrophils from paroxysmal nocturnal hemoglobinurea patients who are deficient in the ability to synthesize GPI linkages. It is up-regulated on granulocytes from polycythemia vera and thalassemia patients. CD177 binds to PECAM-1 on vascular endothelial cells, an interaction which mediates neutrophil adhesion to the vascular wall and neutrophil transmigration. It associates in cis with the Integrin MAC-1 (CD11b/CD18). CD177 also associates in cis with Proteinase 3 (PR3) and is required for cell surface PR3 expression. PR3 is normally found in intracellular vesicles, but once at the cell surface it can serve as an autoimmune target for anti-neutrophil cytoplasmic antibodies (ANCA). The ANCA targeting of CD177-PR3 complexes triggers neutrophil activation and vasculitis. Post-translational: N-glycosylated. A soluble form may also be produced by proteolytic cleavage at the cell surface (shedding).
