生化机理

人类 S-腺苷高半胱氨酸酶（AHCY）是一种细胞质四聚体，每个亚基都有一个紧密结合的 NAD 辅助因子。它是唯一已知的催化 S-腺苷高半胱氨酸（AdoHcy）分解为高半胱氨酸和腺苷的酶。AdoHcy 的水解是一种可逆反应，其平衡有利于 AdoHcy 的形成，但在生理条件下，由于腺苷和同型半胱氨酸的快速清除，水解占主导地位。因此，AHCY 在哺乳动物体内的活性与同型半胱氨酸水平直接相关，而同型半胱氨酸水平是血管疾病的一个独立风险因素。它还通过控制 AdoHcy 的浓度来调节生物跨甲基化，AdoHcy 是所有 S-腺苷-L-蛋氨酸甲基转移酶的强效竞争性抑制剂。人类 AHCY 基因突变会导致 AHCY 缺乏症，引起血浆肌酸激酶、蛋氨酸、S-腺苷蛋氨酸和 AdoHcy 增高、髓鞘化延迟、肌病和精神运动迟滞。
