The optimized SDS-alkali lysis method was used to lyse the cells and remove impurities such as inhibitors efficiently. The silicon-based plasma membrane in the centrifugal adsorption column selectively binds the plasmid DNA in the solution under the state of high salt and low pH, and removes proteins, genomic DNA, RNA and other impurities to the greatest extent. Finally, the pure plasmid DNA was eluted from the silicon-based plasma membrane in the elution buffer of low salt and high DH, and each adsorption column could adsorb up to 50ug of plasmid DNA. Purified plasmid DNA can be directly used in biological experiments such as enzyme digestion, PCR, sequencing, transformation, and transfection of some conventional passaged cells.
