Vibrio Chromogenic Medium (Second Generation)
Name: Vibrio Chromogenic Media
Item number: VIP (HB) 7011-5
Specifications: 1000ml
Uses: It is used for rapid detection of Vibrio parahaemolyticus in food, seafood, patient fecal samples and aquatic products. Vibrio parahaemolyticus is blue-green and has a relatively large colony, Vibrio cholerae and Vibrio vulnificus are purple-red,Vibrio alginolyticusPale yellow or beige; most other bacteria are inhibited. This medium has high specificity and can overcome the false negative results caused by TCBS medium, making it an ideal medium for rapid detection.
Vibrio chromogenic mediumComposition (g/L)
usage
Weigh 18.9g of this product and add 200ml of distilled water, heat to dissolve and keep stirring, boil 3-5 times. When cooled to 45-50 ° C, pour into a sterile plate.
Attention: The surface of the prepared medium should not be too dry.
Operation steps
1. Prepare the sample solution according to national standards, SN standards, FDA standards or other methods.
2. Take 1ml of sample solution and add it to 9ml of SCPB broth or alkaline peptone water, and incubate it at 36 ± 1 ° C for 18-24 hours;
3. Use a 3mm inoculation ring to take 1 ring of bacterial enhancement liquid, scribe and inoculate it on the Vibrio coloring medium, and make 2 plates;
4, 36 ± 1 ℃ culture for 18-24h, Vibrio parahaemolyticus was blue-green, the colony was relatively large; Vibrio cholerae and Vibrio vulnificus purplish red, Vibrio algumolyticus pale yellow or beige, most of the other bacteria were inhibited;
5. Suspected colonies can be inoculated on nutrient agar plates, incubated at 36 ± 1 ° C for 12-16 hours, and a single colony can be selected for a full set of biochemical tests for Vibrio parahaemolyticus.
Quality Control
1. Appearance
The powder of this drying medium is uniform, has good fluidity, and is light yellow. The prepared plate was light pink to yellow solid medium.
2. Microbial testing
Incubate at 36 ± 1 ° C for 18-24h
Limitations of the method
This medium cannot directly identify Vibrio cholerae. The specificity of Vibrio parahaemolyticus can reach 95%, and the accuracy is very high. Sometimes other supplementary tests may be required.
typical features
Typical Vibrio parahaemolyticus appears blue-green and has a relatively large colony, Vibrio cholerae and Vibrio vulnificus appear purplish-red, and Vibrio algumolyticus appears light yellow or beige
storage
The prepared plate can be stored for 2-5 days, and direct light exposure should be avoided. The drying medium should be placed in a dark and dry place, the storage temperature should be 2-8 ° C, and it should be stored away from light.
failure
Dry media should not be used beyond shelf life, clumping, or color changes.
Attention
This medium is for laboratory use only.
